چکیده

Objective: This study investigates the protective effect of butylphthalide on hippocampal mitochondria in epileptic rats. Methods: (1) A rat model of pentylenetetrazol (PTZ)-kindling was used. (2) Enzymatic activity of respiratory chain complexes II and IV was quantified in the hippocampus. (3) Mitochondrial morphology of the hippocampal CA3 region was observed by transmission electron microscopy; (4) Electrical activity of the rat hippocampal CA3 region was recorded with an electrode in vivo. Results: Compared with the epilepsy (EP) group, the butylphthalide (NBP) group showed significantly improved mitochondrial respiratory chain complex IV activity in epileptic hippocampi [Control group (78.2967 ± 7.6304) mmol Cytc/min/mg; EP group (45.3269 ± 11.6556) mmol Cytc/min/mg; and NBP group (72.1153 ± 15.1009) mmol Cytc/min/mg (P < 0.01). In addition, the mitochondrial ultrastructural damage was reduced and the frequency of electrical bursts in the hippocampal CA3 region decreased. Conclusion: Butylphthalide has a protective effect on mitochondrial function in the hippocampus of epileptic rats, thereby reducing the abnormal electrical activity of neurons. It may therefore have potential as an adjuvant in treatment during the active epilepsy phase.

Exercise during adolescence attenuated depressive-like behaviors and hippocampal mitochondrial dysfunction following early life stress in adult male rats

Purpose: In this study, we assumed that treating animals with an antidepressant agents or voluntary running wheel exercise (RW) during adolescence may have protective effects against early life stress (ELS) which can impact on behavior and mitochondrial function. Evidence indicates that ELS has deleterious effects on brain and behavior and increases the risk of mental disorders such as depressi...

The Protective Effect of Voluntary Exercise on the Hippocampal Cerebral Dopamine Neurotrophic Factor Level against Intraventricular Injection of 6-hydroxydopamine in Rats

Background & Aims: The purpose of this research was to study the protective effect of pretreatment with a voluntary exercise on hippocampal level of cerebral dopamine neurotrophic factor (CDNF) after damage induced by intraventricular injection of 6–hydroxydopamine (6-OHDA) in rats. Methods: In this experimental study, 24 Wistar rats were randomly divided into 4 groups of healthy control, healt...

The Effect of 4 Weeks of Aerobic Training on Spatial Learning, Memory Performance and Mitochondrial Dynamics in the Hippocampal Tissue of Old Rats

Background & objectives: Mitochondrial dysfunction is one of the main risk factors for neurological diseases which are associated with aging. On the other hand, aerobic exercise has beneficial effects on the brain health and cognitive function, and also improves mitochondrial dynamics. Therefore, the aim of the present study was to investigate the effect of 4 weeks of aerobic exercise on spatia...

Comparison of the Protective Effects of Curcumin and Nanocurcumin on Acrolein Induced Mitochondrial Toxicity Isolated from Rat Liver

Background and Objectives: Contact with acrolein rapidly reduces intracellular glutathione and its antioxidant capacity and causes mitochondrial dysfunction. The use of some polymeric nanocurcumin increases the stability of curcumin. In this study, the effect of these two, was investigated on acrolein-induced toxicity in mitochondria isolated from rat liver cells.   Methods: In this experiment...
